Charlie Chan visits Treasure Island in San Francisco to investigate a phoney spiritualist who is using mind-reading tricks during seances to obtain blackmail evidence. Along the way, Chan uncovers a hidden secret room and must solve a series of mysterious crimes.

Tana, a voluptuous half-caste girl of the South Seas, falls in love with FBI agent Wally and wishes to marry him and leave the island. Wally, with his pal Jinx, is there to investigate rumored Japanese spy activity. He agrees to marry her if she will help him in his investigations. Through the discovery of a contraband radio set tuned to Japanese reception, Wally learns that the local Commandant is in league with the enemy and is planning an invasion of the island.

Rhythm of the Islands is set in the South Seas, presumably far away from the shooting war. The nonsensical plotline finds hero Tommy (Allan Jones) posing as a native chief. Joan Holton (Jane Frazee), daughter of a millionaire (Ernest Truex), falls in love with Tommy, unaware that he's a charlatan.
